教授,博士生导师。2002 年 6 月毕业于北京师范大学, 获理学学士学位。之后于 2005 年 6 月毕业于中国协和医科大学获医学硕士学位。2009 年 6 月毕业 于北京生命科学研究所,获理学博士学位。后于 2009 年 9 月开始在美国西奈山医学院(Icahn School of Medicine at Mount Sinai, NY, U.S.A.)进行博士后研究至 2015 年底。2015 年获聘中 山大学“百人计划”引进人才,并于 2016 年入选中组部“青年千人计划”引进回国,在中山大学 任职至今。
基于表观调控机理,提高多能干细胞质与量,促进其临床转化。主要成绩:⑴首次发现 TET1 和 TET2 从 DNA 去甲基化的表观层面提高 iPS 细胞诱导效率和质量并阐释其机理(Nature,2013);⑵ 进一步发现新型多能因子 TEX10 可通过超级增强子的表观修饰,促进干细胞多能性(Cell Stem Cell,2015);⑶在此基础上发现超级增强子的表观修饰可被 YY1 与染色质重塑复合物 BAF 协同调 控,活化多能性基因表达(Stem Cell Reports,2018);⑷为以上表观研究建立蛋白组学方法 (Cell Research,2012)。这些研究为提高多能干细胞质与量提供全新表观途径,为其临床转化提供 理论基础。近年共发表 SCI 论著 21 篇,引用超 1300 次,其中以通讯或(共同)一作发表论著 7 篇 (3 篇 IF>15)。
本课题组通过深入研究干细胞的多能性以及体细胞重编程的分子机理,阐明胚胎干细胞的自我 更新、分化及体细胞重编程的发生发展过程,并利用 iPS 细胞建立肿瘤疾病模型,对于干细胞的临 床应用具有重要意义。基于此,实验室正在开展以下几个方面研究:
1、利用蛋白质组学、基因编辑等手段研究多能干细胞维持及分化、体细胞重编程及早期胚胎发 育的表观遗传调控机制; 2、结合相分离和染色体三维结构技术(HiC、HiChIP 等)研究干细胞多能性的表观机制; 3、利用生物信息学技术进行表观组学研究,整合二维的基因组学、转录组学、蛋白组学等数据 和 HiC、HiChIP 等染色质三维数据,进行细胞命运和疾病发生的研究; 4、利用诱导多能干细胞(iPS 细胞)建立人类疾病模型,特别是乳腺癌模型,并利用模型研究 其发病机理,长期目标为筛选药物。
