个人简介

徐涛,中山大学引进中青年杰出人才,教授,博士生导师。在中科院上海生物化学与细胞生物学研究所(现中国科学院分子细胞科学卓越创新中心)获得细胞生物学博士学位。在斯柯里普斯研究所(The Scripps Research Institute)以及美国加州大学旧金山分校戴维•格拉斯通研究所(J. David Gladstone Institutes,University of California, San Francisco)从事博士后研究工作。2018年由中山大学引进全职回国至今。2017年至今已经以独立通讯或第一作者身份在Nature, Journal of Experimental Medicine等杂志发表文章多篇,总引用2000多次。

主要科学贡献

TH17细胞在清除细菌和真菌感染中发挥至关重要的保护性作用,但在一系列的自身免疫性疾病,如多发性硬化病,银屑病,炎症性肠道疾病,风湿性关节炎等都发挥着非常重要的致病性作用,因此研究TH17细胞发育分化具有重要意义。徐涛博士在TH17细胞的发育分化中取得了重要进展,系列工作包括:

  1. 国际上首次揭示谷氨酰胺代谢能够通过影响表观遗传及转录来调控TH17细胞的命运决定(Nature, 2017, 2023);
  2. 证明TCR-Lck/Fyn-STAT3轴在调控TH17细胞分化中的重要作用,提出了TCR信号和细胞因子信号协同作用诱导STAT3的磷酸化,进而促进TH17细胞命运决定这一全新的理论模型(Journal of Experimental Medicine,2024)。

Cell Metabolism, Nature Review Immunology, Cell Research等国内外顶刊为这些工作发表了专评。相关工作已被Nature, Cell, Science, Immunity, Nature Immunology, Nature Biotechnology, Nature Review Immunology, Annual Review Immunology,Nature Review Drug discovery等顶级刊物多次正面引用。这些工作对于推动TH17细胞的发育分化以及TCR信号和细胞内代谢在T细胞命运决定中的作用提供了全新的认识,并为靶向TH17细胞的药物研发提供了全新的靶点。

目前研究方向

肿瘤免疫疗法是近年来生物医学领域内最重大的突破。Science杂志将其列为2013年度最重要的科学突破。2018年诺贝尔生理医学奖授予了在肿瘤免疫疗法,特别是免疫检查点疗法中做出巨大贡献的两位科学家James Allison 和Tasuku Honjo。与传统的化疗药相比,肿瘤免疫疗法具有治愈后维持时间长(可以长达10年以上)和疾病不再复发(disease free survival)的优势。目前临床上使用的免疫检查点阻断疗法主要是通过恢复抗原特异性T细胞的功能,从而使其更好的发挥肿瘤杀伤能力。

实验室目前致力于:

1)抗原特异性T细胞和其识别的多肽抗原的快速识别和鉴定;

2)研究抗原特异性T细胞在肿瘤内部如何发育分化,从而为如何提高此类T细胞的肿瘤杀伤功能提供新的靶标;同时也关注在其它疾病(如自身免疫性疾病)中,抗原特异性T细胞在体内如何发育分化,从而为更精准靶向致病性T细胞提供新靶点;

3)肿瘤细胞是如何逃脱免疫监视。

同时我们也将着力于将这些基础研究的成果转化为临床应用。
